5-HTP (5-Hydroxytryptophan)

5-HTP (5-hydroxytryptophan), a metabolite of tryptophan, has more consistent effects than tryptophan in improving sleep. It is one step closer to the manufacture of serotonin, a neurotransmitter that plays a key role in initiating sleep. And since melatonin is synthesized from serotonin, 5-HTP supplementation indirectly increases melatonin levels. Clinical studies show that 5-HTP is considerably more effective than L-tryptophan.

One of the key benefits of 5-HTP noted in human clinical trials is its ability to increase REM sleep (typically by about 25%) while increasing deep sleep stages 3 and 4 without lengthening total sleep time. The sleep stages that are reduced to compensate for the increases are non-REM stages 1 and 2 — the least important stages.1,2

Magnesium is very safe at recommended levels. However, caution is advised for people with severe kidney impairment, for anyone taking tetracycline or fluoroquinolone antibiotics (space doses several hours apart, since magnesium can reduce absorption of these antibiotic classes), and for anyone on blood pressure medication — particularly calcium channel blockers like amlodipine or diltiazem. Magnesium acts as a natural calcium channel blocker itself, and the two effects can stack, dropping blood pressure lower than intended.

L-Glycine

Glycine is one of the most intriguing sleep aids. Glycine is an amino acid, but it is also a neurotransmitter with measurable effects on sleep physiology. Glycine doesn't "knock you out" like melatonin or herbal sedatives. Instead, it fine-tunes the body's natural sleep physiology.

As a neurotransmitter, glycine calms neural activity in the spinal cord and brainstem. One of the effects of taking glycine is that it lowers core body temperature by increasing blood flow to the skin, mimicking one of the body's natural signals to initiate sleep. This lowering of nighttime body temperature appears to help synchronize circadian rhythms and sleep onset.6,7

In a human double-blind, crossover trial in subjects who complained of poor sleep, taking 3 g glycine before bedtime improved subjective sleep quality, shortened time to fall asleep, and reduced fatigue and sleepiness the next day compared with the placebo. In another study, taking 3 g of glycine before bedtime led to shortened sleep latency, increased time in slow-wave sleep (deep sleep), and improved next-day performance on cognitive tests.8,9

Glycine, like magnesium, is very safe, suitable for children, easy to use, widely available, and relatively inexpensive.

L-Theanine

L-theanine is a unique amino acid found almost exclusively in tea (Camellia sinensis). Clinical studies have demonstrated that L-theanine reduces stress, improves the quality of sleep, diminishes normal premenstrual discomfort, heightens mental acuity, and reduces the negative side effects of caffeine.10

L-theanine is a great consideration for children at a dosage of 200 mg at bedtime. In adults, the typical dosage of 200 mg does not act as a sedative, but it does significantly improve sleep quality — a gentle way to improve sleep. For a noticeable sedative effect in adults, a higher single dosage of 300 to 600 mg is required.

GABA

Gamma-aminobutyric acid (GABA) is a major neurotransmitter that is abundantly and widely distributed throughout the central nervous system (CNS). GABA functions as one of the brain's most important regulators of proper function. Insufficient levels of GABA may produce poor sleep quality, stress, and low mood.

Taking a dietary supplement form of GABA is an obvious method to try to boost GABA effects in the brain. But it is not that simple. First, the form of GABA is important. Synthetic GABA has not shown the same benefits as natural forms of GABA.11,12

Natural forms of GABA have been shown to promote improved sleep, especially with continued use.12 In one study with PharmaGABA, 38 senior-aged subjects were given 100 mg of PharmaGABA or a placebo for four weeks.13 In the PharmaGABA group, people experienced getting to sleep more easily, reduced nighttime awakenings, less drowsiness in the morning, and improved recovery, along with a reduced frequency of nighttime urination.

Even a single administration of PharmaGABA has been shown to improve sleep. Another double-blind study assessed brain wave patterns through electroencephalography (EEG) after subjects took 100 mg of PharmaGABA or a placebo.14 The results demonstrated that PharmaGABA significantly shortened the time required to get to sleep by five minutes and increased the time of quality sleep compared to the placebo, with subjects reporting improved sleep and higher energy scores upon awakening.

In a study with a GABA product derived from fermented barley (Sanwa GABA), men and women between the ages of 39 and 59 were given either 100 mg of Sanwa GABA or a placebo 30 to 60 minutes before bedtime. Results showed GABA significantly increased deep sleep in subjects over 44 years old and/or those experiencing strong psychological stress or fatigue.15 In another double-blind study with Sanwa GABA, 62 subjects dealing with sleep problems, stress, and fatigue experienced significant improvements in sleep quality, subjective fatigue ratings, and feelings of vigor and vitality.16 

Lactiplantibacillus plantarum Lp815®, a GABA-Producing Psychobiotic

Lactiplantibacillus plantarum Lp815® is a novel psychobiotic—a probiotic strain selected for its ability to influence communication between the digestive tract and the brain. Lp815® is distinguished by its capacity to produce gamma-aminobutyric acid, or GABA, under conditions similar to those found in the human intestinal tract.17 

Unlike a single oral dose of GABA, Lp815® may provide ongoing GABA production within the gastrointestinal tract, potentially supporting the gut-brain axis over an extended period. The benefits of Lp815® have been demonstrated in a randomized, double-blind, placebo-controlled human clinical trial involving 139 adults with sleep disturbance.18 Participants received either 5 billion CFU of Lp815® or placebo daily for six weeks. By the end of the study, the Lp815® group had significantly improved sleep scores compared to the placebo group. A clinically meaningful improvement of at least four points occurred in 77.3% of participants receiving Lp815®, compared with 57.8% receiving placebo. Lp815® improved both difficulty falling asleep and difficulty staying asleep. Objective sleep measurements collected with wearable sleep-monitoring devices also showed increases in total sleep duration and deep sleep duration compared with placebo. Everyday stress scores improved as well, with particularly notable benefits observed among women.

Valerian

In terms of herbal medicine, there is no question that valerian (Valeriana officinalis) is the most popular and well-studied sleep aid. Detailed clinical studies have shown that valerian improves sleep quality, reduces the time required to fall asleep, and promotes restful sleep throughout the night,19 all without producing a "hangover feeling" in the morning.

In the latest study, researchers examined 80 adults with mild sleep problems who took either valerian extract (200 mg with 2% total valerenic acid) or a placebo for 8 weeks. The results showed that people taking valerian extract experienced better overall sleep quality, fell asleep faster, slept longer, and had more efficient sleep compared to those on a placebo. Improvements appeared as early as two weeks and continued through the full study period. Valerian also reduced feelings of stress and daytime drowsiness and helped participants feel more refreshed in the morning. Advanced sleep testing confirmed longer and better sleep in the valerian group, and no safety concerns were observed.20

As a mild sedative, for best results, take valerian extract (0.8% to 2% valerenic acid) at a dosage of 200 to 400 mg thirty to forty-five minutes before retiring. If morning sleepiness occurs, reduce the dosage. If the dosage was not effective, be sure to eliminate factors that disrupt sleep — such as caffeine and alcohol — before increasing it.

One rare but documented caution: long-term, high-dose valerian use has been linked to a small number of reversible liver-injury cases (NIH's LiverTox database lists it as a "probable rare cause" of clinically apparent liver injury, typically resolving after stopping use). This isn't a reason to avoid short-term, standard-dose use, but anyone with existing liver disease, or using valerian continuously for more than a few months, should mention it to a doctor.21

Lemon Balm (Melissa officinalis) 

Lemon balm has long been used as a traditional sleep aid  for its ability to lessen the emotional and mental factors that often interfere with the ability to relax at night.22 The best clinical validation for lemon balm is the phytosome form known as Relissa® that complexes lemon balm’s naturally occurring active compounds with phospholipids to improve their absorption and biological availability compared with conventional lemon balm preparations.

In one randomized, double-blind, placebo-controlled clinical trial, adults experiencing emotional distress and poor sleep received 400 mg of Relissa® daily for three weeks. Compared with placebo, supplementation produced significant improvements in overall sleep quality, mental well-being, mood, and perceived stress.23

In another double-blind, placebo-controlled study in adults with poor sleep quality, Relissa® significantly improved several important measures of sleep. These included sleep quality, the amount of time participants slept, sleep efficiency, and the ability to remain asleep without prolonged nighttime awakenings. Overall, 87% of participants reported improved sleep quality while taking the lemon balm preparation, compared with 30% while taking placebo.24

Apocynum venetum Leaf Extract

Apocynum venetum leaf, also known as rafuma or luobuma has a long history of use as a calming tea. Newer preparations standardized to provide their key active flavonoids have been developed to deliver precise, consistent levels of these biologically active compounds.25

Apocynum leaf extract supports sleep partly by helping the body adapt to psychological stress and maintain a more balanced emotional state. This is important because stress, worry, and heightened mental alertness are among the most common reasons people have difficulty falling asleep or experience fragmented, nonrestorative sleep.

In a randomized, double-blind, placebo-controlled crossover study, adults who consumed 50 mg of an apocynum extract known as Venetron® experienced measurable improvements in sleep physiology. Brain wave testing showed that the extract increased the deeper stages of sleep by approximately 7.6%.26 

Additional human clinical research has evaluated Venetron®, showing that daily supplementation was associated with improvements in perceived stress and measures of sleep quality, supporting its role as both a calming botanical and a natural sleep aid.27

Rather than acting as a conventional sedative, apocynum extract appears to promote sleep by supporting relaxation, emotional resilience, and a healthier transition from daytime stress to nighttime rest.

Passion Flower (Passiflora incarnata)

Another plant with a long history as a sleep aid is passion flower. It contains several bioactive constituents — including flavonoids and alkaloids (such as harman, harmaline) — that interact with the central nervous system. Preclinical evidence suggests it enhances GABA (gamma-aminobutyric acid) activity, which may explain its calming and sedative effects.

Several human trials have assessed the calming effects of passionflower before surgical procedures, showing it may significantly reduce preoperative stress without side effects. Human studies have also shown that both passionflower tea and various extracts, given for a week or two, led to significant improvements in subjective sleep quality.28-30

The recommended dosage as a sedative, 45 minutes before retiring, for the various forms of passion flower is:

  • Dried herb (or as tea): 4–8 g
  • Fluid extract (1:1): 2–4 ml (0.5–1 tsp)
  • Dry powdered extract (2.6% flavonoids): 300–450 mg

Interactions and Who Should Be Careful

  • 5-HTP + antidepressants (SSRIs, SNRIs, MAOIs) or migraine/pain medications (tramadol, triptans): Do not combine. 5-HTP raises serotonin directly, and stacking it with a drug that already increases serotonin activity has caused documented cases of serotonin syndrome — a medical emergency with symptoms including agitation, rapid heart rate, sweating, and muscle spasms.²⁴ If you're on any antidepressant, talk to your prescriber before trying 5-HTP, and don't stop or start either one without medical guidance.
  • Extracts of valerian, lemon balm, apocynum, and passion flower + benzodiazepines, or other sedatives: All these herbs contain compounds that may produce over-sedation when combined with prescription sleeping pills. 
  • Magnesium + certain antibiotics, severe kidney impairment, or blood pressure medication: Magnesium can reduce absorption of tetracycline and fluoroquinolone antibiotics if taken too close together — space doses a few hours apart.
